Returning to Dagobah to complete his training, an ailing Yoda told Luke the terrible truth that Vader was his father, Anakin Skywalker. Rather than destroy his father, Luke sought to bring him back into the light.
Yoda. Yoda's knowledge of Luke Skywalker's true parentage was also confirmed in Return of the Jedi. As with Obi-Wan, Yoda believed that Anakin's fall to the dark side made him unsalvageable, with the Darth Vader persona being his only identity following his ascension to Sith Lordship.
Jedi Master Yoda's Vader confirmation is the main reason he appears in Return of the Jedi, as explained by George Lucas in the 2004 DVD release. Yoda was not originally in the script for Return of the Jedi, but George Lucas included the character to confirm that Vader really is Luke's father.
Master Yoda knew that Darth Vader was once Anakin Skywalker- the boy he sensed too much fear in to train way back in Star Wars: The Phantom Menace.
The Obi-Wan Kenobi of it all
We know exactly when Vader discovered that Luke was his son, and we know when he figured out that Leia was connected to him as well.
Anakin, who has since become the Sith Lord Darth Vader, is unaware of his children's birth. The twins soon became part of the Rebel Alliance following the deaths of their adoptive families at the hands of the Empire.
Once more, Darth Vader learns the name in the opening scene of an issue (in this case Vader's Quest #1) while torturing a Rebel, though in this case, he only learns Luke's surname of Skywalker, making the Sith Lord's discovery in the classic Marvel series Luke's first name.
This is confirmed later in the movie when Obi-Wan informs Padme that Anakin killed them. Both are torn apart with hurt and confusion, trying to make sense of something so terrible.
In the material around the film from the Legends timeline, it's established that by the end of Star Wars: Episode III - Revenge of the Sith, Kenobi was one of the characters who knew Anakin's Sith name was Darth Vader. However, what he didn't know was that he was still alive after Mustafar.

R2-D2 was present when Padmé gave birth to Leia and Luke, so he knew Luke was Anakin's son – but what he didn't know was that Anakin became Darth Vader. In addition to that, Anakin's fall was surely a painful event in R2-D2's life, one that he definitely wouldn't like to remember or even talk about.
Obi-Wan wanted to protect Luke
One theory which explains why Obi-Wan lied to Luke about his father is that he wanted to protect Luke from the truth.

If you are asking about Vader post-Mustafar, then no, she doesn't. Padmé doesn't seem to know Obi-wan cut off his limbs and left him to burn alive, Obi-wan didn't believe Anakin to be alive and Anakin's surgeries were still happening as Padmé died so there was no way for her to know.
Despite seeing Vader as an idol, Kylo conveniently ignored his grandfather's redemption. He was well aware of what happened, and it's likely Luke himself who told Ben about the events of Star Wars: Return of the Jedi.
Anakin does not have even the slightest bit of remorse. By the end, he feels nothing but anger. Although he feels regret for killing Padme, he still helps the evil Emperor. The film ends with Anakins standing side by side with the man on whose behalf he has committed all his crimes.
