So to put it bluntly, John Winchester is Ben's real father, making Ben actually Dean's little brother. Lisa says how she “has a type leather jacket, couple of scars, no mailing address” and she met the dude in some biker bar. As we all know, John has been with another woman that had resulted in a child.
In a 2008 interview, Eric Kripke revealed that Dean is not the father of Ben.
Lisa Braeden is a single mother of one, who had a fling with Dean Winchester back in 1999, whilst he was on a hunt in the vicinity. She has a young boy named Ben who shares similarities to Dean, although she has categorically stated that he is not his child.

Lydia met Dean Winchester, along with Castiel at a bar. She shared a conversation with the Winchester and then went to her house where the two enjoyed a night together. Unknown to Dean, Lydia was impregnated with his child and in just a couple of hours, Lydia was already nine months pregnant.
Due to Jack being the son of Lucifer, Dean originally viewed Jack as a threat that had to be stopped from being born. After Jack was born, with Sam unwilling to kill him, Dean would keep an eye on Jack, threatening to kill him if Jack ever turned evil. Jack in turn sought Dean's approval and affection.
Adam Millagan is a supporting character in Supernatural. He is the younger half-brother of Sam and Dean Winchester and the son of John Winchester. Do Dean & Sam Have Children? This one is a bit of a doozy, but the answer is yes. Dean accidentally conceived a child with Lydia, an Amazon who had a one-night stand with the Winchester brother in order to have a child. Jack Kline (b. May 18, 2017) is the son of Lucifer and Kelly Kline. He is the first and only known Nephilim to be sired by an Archangel. Jack was taken in by Sam and Dean Winchester who would teach him how to master his powers.
After the second death of Lucifer and the betrayal and death of Michael, Jack absorbs God's power and becomes the new God, the effort to turn him into a cosmic bomb having also made Jack a power vacuum that sucked up all of the power around him, including from the fights with Michael, Lucifer and Chuck.

Emma was an Amazon conceived by Lydia and Dean during a one-night stand, unbeknownst to the latter. After trying to kill Dean, Sam shot her, killing her instantly.
