Dean tells Lisa "you knew what you signed up for". Lisa replies, "I did, but I didn't expect Sam to come back". She knew their relationship was "over the moment Sam walked in the door", and tells Dean how weird his and Sam's relationship is. With Sam, Dean can never be happy.
At the motel, however, Dean drives off to visit Lisa Braeden. He tells her that, in an ideal world, he would want nothing more than to live in peace with her and her son Ben, but knows that this is impossible.

Anna surmises that Castiel and Uriel have been ordered to kill her as punishment for the serious angelic crime of falling. To celebrate her last night on Earth, she has sex with Dean in his car; afterward, when Dean is asleep, he is visited in a dream by Uriel, who reveals that he has Anna's grace.
The line ''I love you'', was to be directed at Cas's character and Jensen put a stop to it. Dean had no reason to say something like that to Cas, of all people. This is the response I get. Or, so Jensen just wants to stay true to Dean's character.
Throughout the second season, Jo appears to have romantic feelings for Dean, but Dean does not have feelings for her, it seems to him that she is like a sister to him.
As Sam prepares to sacrifice himself to Lucifer to stop the Apocalypse, he urges Dean to seek out Lisa after his death. Dean does this, and lives with Lisa and Ben for a year. He has a monogamous relationship with Lisa, and takes on a paternal role with Ben.
Instead, Dean performs an exorcism on Lisa to get the demon out of her. In order to stop Dean from exorcising her, the demon stabs Lisa in the stomach with a tool, leaving Lisa mortally wounded. As a result, if Dean completes the exorcism, Lisa will die.
Cassandra Masters is the daughter of Castiel and Meg Masters and the only known Nephalem to exist, which is a hybrid between a union of an angel and demon, even being part human due of Castiel and Meg's human vessels. Cassandra acts as one of the leading main characters with Sam and Dean.
Lydia met Dean Winchester, along with Castiel at a bar. She shared a conversation with the Winchester and then went to her house where the two enjoyed a night together. Unknown to Dean, Lydia was impregnated with his child and in just a couple of hours, Lydia was already nine months pregnant.
This is reconfirmed in the Supernatural finale when Sam's soul ascended into Heaven and materialized next to Dean's soul, making him the first person he interacted with in the afterlife. The brothers are unique in that they are the only soulmates who are family rather than spouses.

Eventually, it turns out that Amara was only interested in Dean because she did not want to be alone after taking revenge on God. She has even stated that she has loved God more than he could ever imagine.
