In truth, he is desperately looking for a rare flower blue spider lily. Even after centuries, Muzan never gave up in his search for it. However, the flower is so rare that he never even got a hint as to where to find it. This flower is significant to the series as it appears in the outro of Season 1.
The Blue Spider Lily is an important ingredient in the medication prescribed to Muzan during his mortal days. It is the very medicine that allowed him to become a demon. Since Muzan isn't the one who created the medicine and he ends up killing his doctor, he has been in constant search for it.
The spider lily is part of this rich flower language and often is used to foreshadow a coming death. Red spider lilies can be seen in passing in one of my favorite shows, Kimetsu no Yaiba (鬼滅の刃) or Demon Slayer in English, in the ending credits.
Why does Muzan want the Blue Spider Lily so much? As mentioned earlier, the Blue Spider Lily was the final ingredient to complete his treatment. On completion, he will finally be able to conquer the Sun, the only weakness demons have.
Nezuko's Sunlight Immunity in Demon Slayer is Tied to the Blue Spider Lily. Nezuko having eaten the Blue Spider Lily at some point could explain her immunity to sunlight, and Demon Slayer: Kimetsu no Yaiba has already worked to justify that.

Why did Muzan cry at the end? The manga explains that Muzan found Tamayo four centuries ago when she was begging him to cure her of her illness. He turned her into a demon without telling her, and she went crazy. She mercilessly massacred her own family and after she was done, she cried for her loss.
Since Nezuko is from a Sun Breathing lineage, she can survive in daylight. After the final battle against Hantengu concludes, she can step into daylight, and many theories are available about the phenomenon. One of the theories is that her abstinence from consuming human blood is what saved her.

Curse of Kibutsuji
This curse besotted any who partake of Muzan's blood must be careful. The curse prevents Muzan's name from being spoken aloud, and any who do will meet a horrible demise. One demon, Susamaru, faced this reality when battling Tamayo.
Despite being the unstoppable force of nature in Demon Slayer, Muzan is still incapable of conquering the Sun.
