The Blue Spider Lily is an important ingredient in the medication prescribed to Muzan during his mortal days. It is the very medicine that allowed him to become a demon. Since Muzan isn't the one who created the medicine and he ends up killing his doctor, he has been in constant search for it.
Why does Muzan want the Blue Spider Lily so much? As mentioned earlier, the Blue Spider Lily was the final ingredient to complete his treatment. On completion, he will finally be able to conquer the Sun, the only weakness demons have.
A millennium ago, during the Heian Era, Muzan was turned into a demon during an experimental treatment trying to cure his terminal illness, and his goal since then was to live without the fear of death and become truly eternal.
The spider lily is part of this rich flower language and often is used to foreshadow a coming death. Red spider lilies can be seen in passing in one of my favorite shows, Kimetsu no Yaiba (鬼滅の刃) or Demon Slayer in English, in the ending credits.
In some cases, depending on the climate, the Blue Spider Lily doesn't even bloom at all. This explains why Muzan Kibutsuji never found it despite searching all over Japan for centuries: he was hindered by his inability to go out during the day.

In a nutshell, Muzan, being the progenitor of demons, is not scared of Doma. However, he is irritated by his inability to decipher Doma's thoughts and feelings, which is the ultimate control he has over all of his creations.
