The ability to actually speak Parseltongue — not just imitate it as Ron Weasley did — is considered an attribute of a Dark Wizard, which is partly due to the fact that both Salazar Slytherin and Lord Voldemort possessed this ability. No explanation is given in any of the novels. However, when J. K. Rowling was asked the same question at an interview, she said Dumbledore had studied Parseltongue and could therefore recognize and understand it. That shouldn't be compared to Harry or Voldemort's Parseltongue abilities, though.
