The most common story involves him falling into a tank of chemical waste that bleaches his skin white and turns his hair green and lips bright red; the resulting disfigurement drives him insane. The antithesis of Batman in personality and appearance, the Joker is considered by critics to be his perfect adversary.
A luscious head of green hair is usually one of Joker's defining traits, but The Batman's deleted scene reveals Keoghan's head covered in only small green tufts sporadically dotted across his scalp. A view from behind even shows Joker verging on the edge of baldness.
For Joker's iconic green hair, it was production designer Mark Friedberg who suggested what became the final look. “He said it should be a broccoli green,” says Georgiou.
Her hair colors are because DC wanted to pay homage to her classic look. There isn't really a in story explanation for why her hair didn't turn green. In DC Rebirth, she still has pale white skin like Joker, but her hair is blonde with parts of it dyed. In Batman TAS, and the Pre-52, she wore makeup.

This haircut is called an undercut.
At times, the Jokers will each be colored to match the colors used for suits; e.g., there will be a red Joker and a black Joker. In games where the Jokers may need to be compared, the red, full-color, or larger-graphic Joker usually outranks the black, monochrome, or smaller-graphic one.

Pure green was off-limits to us (as well as the name “Joker”), a decision from high-up as they wanted to reserve these for films,” Monaghan explained.
In the end, we never find out, but what we do know is the Joker's a lunatic who smears clown paint on his face to frighten people—that's the look this Joker needed, and ultimately it's the look that Caglione Jr was able to create.
His face is indeed white like clowns' faces, but this is because he literally fell into a vat of acid, while real clowns (and the Red Triangle Gang) use face paint. He eschews the clown color scheme and doesn't wear a frilly collar or red nose, instead opting for a sleek purple and green suit.

However, it was probable that the chemicals that caused the Titan Disease (and maybe chemicals already in the clown's blood) caused Joker's blood to mutate creating the infection.
Following her introduction to the DC Universe in 1999, Harley Quinn was depicted as the sidekick and lover of the Joker as well as the criminal associate and best friend of fellow supervillain Poison Ivy.

His hair was short in The Phantom Menace, but in Attack of the Clones he's wearing something of a modified mullet, with a dinner party in the front and a real rager in the back. “We called it the sort of Jedi mullet,” McGregor said.
In Joker's case, pseudobulbar affect probably occurred secondary to severe traumatic brain injury (TBI). A number of studies have established that TBI increases the risk of mood disorders, personality changes and substance use disorders.
The psychopathology Arthur exhibits is unclear, preventing diagnosis of psychotic disorder or schizophrenia; the unusual combination of symptoms suggests a complex mix of features of certain personality traits, namely psychopathy and narcissism (he meets DSM-5 criteria for narcissistic personality disorder).
Harley's ADHD is apparent in her hyperactive nature and temper tantrums.
