Thanos didn't know the location of the soul stone. He knew the location of the other five, but he was waiting until he knew the location of all six before he made a major move to collect them.
By the events of 2012's The Avengers, Thanos already had the Mind Stone, so it's safe to say that he had been looking for the gems for around 15-20 years before he successfully acquired all of them in Avengers: Infinity War.
Thanos is a very unique character. In his own way, he is very wise, and he is very patient. He sits patiently, because he knows the Avengers will come to him, given he just attacked the base. He's ready for a fight, but is in no hurry for it.
However, unlike Red Skull, he didn't manipulate the Stones with his bare hands; instead, he used the Infinity Gauntlet to mount them in and channel their power in a more secure manner. It's possible that Thanos' use of the Infinity Gauntlet protected him from being punished in the same way that Red Skull was.
He simply wanted him dead at all costs because he was his biggest threat. As per the user, the director of Avengers confirmed that Thanos was most scared of Iron Man. “Not because of his strength, not because of his power, not because of his power, but because of his suit,” he added.
Thanos vs MCU Hulk
This short battle was shown in the aforementioned Infinity War's opening scene. The Mad Titan was easily able to dispatch the Hulk without even using the only Infinity Stone he had available at the time: The Power Stone.
While he might have been able to defeat him, the Hulk could inflict some serious damage to Thanos and could conceivably kill him if given the chance. That threat to his life would be a completely unnecessary risk for Thanos to take without the absolute power of the Infinity Stones.

What is the seventh Infinity Stone? Earlier this month, Marvel Comics confirmed the existence of a seventh Infinity Stone, the Black Stone. Its existence was confirmed in Thanos: Death Notes #1 – Dead by Rumor, a new comic book series one-shot featuring Thanos that kicked off on November 30.
Thor loses his eye during the earlier parts of the final battle sequence with this goddess. She casually and effortless beats him around the throne room, and then the moment happens. Hela strikes Thor across the face with her sword, literally slicing the eye from out his head right then and there. Brutal.
Of course, the Power Stone, Reality Stone, and other Infinity Stones are nothing to scoff at, but the Soul Stone's collective abilities make it the one gem that is the strongest overall.
No, he doesn't. He doesn't fear Odin to the point even a spear-wielding All-Father respects him as an adversary worth saluting, considering him as a foe he's never fought in eons.
Thanos was scared of the ancient one, Odin, and Ego. That's why Thanos didn't show up until infinity war. This is why Thanos waited for them to die before he striked.
Zeus was confirmed to be apart of the MCU in the newest Marvel movie still in theatres today. As long as Thanos doesn't disturb Zeus' possessions of his, nor kill his more personally close gods he would not intervene as he simply does not care for mortal deaths or gods that he does not have a close relationship with.
That villain is, of course, Kang the Conqueror, who's set to be the Avengers' next opponent in the upcoming Avengers: The Kang Dynasty. And while Thanos is certainly a hard act to follow, Kang may be a far greater threat than the Mad Titan ever was.
Sure, Hulk's had rough fights before, but Thanos just completely dominated him, and the theory was that it had intimidated the big green guy into inaction.
Thanos has, in the comics, repeatedly beaten Hulk with and without the Infinity Gauntlet. And he had also beaten him in the Marvel Cinematic Universe when he smashed him in a fight without using the Infinity Gauntlet.
The theory posits Ego, Odin, and The Ancient One as three of Thanos' greatest fears, and he waited until all of them were dead to make his move. This would make sense, as all of these characters were massively powerful and had strong ties to the Avengers, which meant they could be called in for backup at any time.
The Asgardian Son of Odin was extremely close to ending Thanos at the end of Avengers: Infinity War. After gaining the powerful weapon Stormbreaker, Thor becomes the wielder of the most powerful weapon in the universe.
And in the MCU without infinity stones, Thanos' abilities are limited only by physical strength and durability. On the other hand, comics Thanos was psychologically weaker & couldn't beat up the Hulk, so it was about even.
Why didn t Captain Marvel fly through Thanos? She isn't powerful enough to do that. In a one on one fight, even with out the Infinity gems, Thanos is just too much for her.
Thanos didn't lift Mjolnir, Captain America did
With Mjolnir's worthiness enchantment resulted in Thor being able to lift the hammer only if he is considered to be worthy, but likewise, the enchantment is not discriminatory in nature, and anyone who is deemed worthy is able to lift the hammer.
Because Thanos is more than durable enough to tank her attacks. His best durability feat is enduring a power surge of unparalleled cosmic proportions from snapping the Infinity Gauntlet. When Thanos snapped his fingers, Earth became ground zero for a power surge of ridiculously cosmic proportions.
