John and Jamie have a complicated relationship throughout the series. While John did say he loves Jamie, he doesn't seem to show it, ever. One could even say John is hard on him at all times. By how he treats the rest of his children, it's easy to conclude that John favors Beth and Kayce over Jamie.
There are plenty of details to suggest that “Yellowstone's” smartest Dutton probably had some sort of idea that Jamie was adopted. However, the moment was not a part of the onscreen story. But Beth likely didn't know that Garrett Randall was Jamie's biological father until season four – after the attacks.

When Beth was a teenager, she got pregnant. She sought help from Jamie, who assisted her efforts to get an abortion. But, in order to keep the pregnancy from leaking to the press and embarrassing the family, Jamie took Beth to a free clinic on a Reservation, where sterilization was mandatory with abortions.
Rip Wheeler and Jamie aren't related by blood, and if we're being honest, there's not much that connects them except for the Dutton name. One connection is that they both swore to John Dutton to protect the family and its reputation.

The second son of James and Margaret Dutton, Spencer was born some time after the family settled in Montana. Spencer also served in WWI and, like his father, remained haunted by his experiences on the battlefield. Rather than returning to the family's ranch after the war, he became a big game hunter in Africa.
As fans gained more knowledge about John Dutton's (played by Kevin Costner) lineage, they appear to have exposed a major inaccuracy with his great-great uncle, Spencer Dutton (Brandon Sklenar).
The 1923 patriarch of the Dutton's and owner of the ranch, Jacob is the brother of 1883's James Dutton. Jacob took over the Yellowstone Dutton ranch after James and his wife, Margaret died, taking in the couple's two surviving children as well.
