According to J.K. Rowling's Pottermore entry on Remus, his Patronus was a normal wolf, not a werewolf, due to their non-threatening and family-orientated nature.
When James learned what Sirius did, he stopped Snape before he reached the Shrieking Shack, saving his life. Snape, however, had seen Lupin in werewolf form and was sworn to secrecy by Dumbledore.
And Remus Lupin was bitten when he was very young, long before he even knew that there was something called an Animagus. So he couldn't have been an Animagus before he got bitten. And being a werewolf, it was no longer possible.

Sirius Black had a lot of love in his life. Mainly for his best friends but still. One of the greatest loves came not from his connection to James Potter but rather with his relationship to Remus Lupin.
Snape thought Sirius wanted to murder Harry. Snape thought Lupin was helping Sirius to sneak into Hogwarts and then yes murder Harry. So of course Snape wanted Lupin out the castle, based on the information he had he believed he was saving Harry's life.

But Lupin was actually in despair, convinced that he could not run the risk of marrying and passing on his lycanthropy to a child. Tonks, convinced that Lupin would never spend time with her again, became desperately unhappy.
Remus was only four when he was bitten, and spent a difficult childhood coming to terms with what had happened. Even worse, Greyback only targeted Remus because his father, Lyall, had insulted werewolves.
While in school together, Sirius once tricked Snape into almost entering the Shrieking Shack while Lupin was there, transformed into a werewolf. James realised the danger and stopped Snape, saving his life; this is the incident Dumbledore referred to at the end of the first book.
Ron, Harry, and Hermione assume she is upset over Sirius' death, and Harry even suggests that Tonks may have been in love with him, which is why she's so crestfallen. It is only at the end of the book that we find that it is Lupin, rather than Sirius, who is the object of her affections.
Also during his time in school Remus began to form an attraction and fall in love with Alice Fortescue. Remus chose to ignore his feelings in the beginning because he felt he was too dangerous for her. Later on in life he falls in love again and marries Nymphadora Tonks.
