That issue finally answered the question: Richard and Mary Parker were murdered by Albert Malik, who was one of Johann Schmidt's successors to the persona of Red Skull. In The Amazing Spider-Man #365 (August 1992), Spider-Man's 30th anniversary, they reappeared.
While traveling on the plane, Richard and Brock, along with their wives, perished as the plane crashed. Brock's son, Eddie Brock Jr., and Peter Parker ended up creating the Venom Symbiote. In another plot in the Ultimate Marvel universe, the Hulk is responsible for the untimely death of Peter's parents.
Richard Parker is actually still alive. Richard Parker was experimenting on Peter Parker as a child, which is why Peter is able to apparently control his mutation, unlike The Lizard. Peter's real father is Norman Osborn.
The Skull and his goons ultimately sniff out the ruse, but Richard and Mary are able to survive the plane crash that allegedly killed them by parachuting out before impact (Richard was trained to do this, 'natch).
While on a mission to investigate Albert Malik, the third Red Skull, they posed as traitors and double agents to infiltrate his criminal organization in Algeria, ultimately being discovered. Malik had an assassin kill the two by sabotaging their airplane and causing it to crash.
The carjacker attempts to shoot him, but ends up tripping and falling to his death. In Spider-Man 3, the Parker family learn the carjacker, now identified as Dennis Carradine, was not Ben's killer. Instead, it was Carradine's partner, Flint Marko, who accidentally shot Ben when the former startled him.

Richard Parker is the Bengal tiger that symbolizes God or faith through love and fear, and also survival. He symbolizes being an imaginary friend as well. However, it can also be argued that Richard Parker symbolizes that he and Pi are one in the same.
A lot of comments on here claim Richard did not look back because he was an animal. This is true, but there is more to it than that. The tiger did not look back because although in that form his job was finished, he would always be in Pi's life in whatever form of life he took.
