For all his powers, Spider-Man is still just a man. He is covered in mortal flesh that can bruise and bleed. He doesn't possess any kind of invulnerability, like some of his superhero buddies. He relies on his speed and agility to stay out of the way of lethal hits.
The red-sun radiation negates Superman's invulnerability in regards to Spider-Man, allowing Spider-Man's punches to hurt Superman until the radiation dissipates, and Superman is again invulnerable.

There are few heroes the Hulk hates the way he does Spider-Man. Spider-Man can't hurt the Hulk on his best day, but he's so annoying that the Hulk can't help but hate him. Spider-Man is lucky he's so agile because the Hulk would love nothing more than to smash him like a bug.
