Strange willingly gave the Time Stone to Thanos to save Tony Stark's life. He saw 40,000,605 possible futures and outcomes of the battle against Thanos. He knew that Tony must stay alive so they could come victorious ultimately and restore the universe as it was before Thanos's snap.

Doctor Strange's third eye (the Eye of Agamotto) in the comics, according to Bustle, represents wisdom of the sorcerer Agamotto and his Vishanti. The Eye allows him to see the true intentions of his enemies and break down all magical illusions.

In Endgame, the significance of Strange raising that finger is that he's telling Tony that this WAS the one possibility, everything was going the way the saw it. This also means that he saw Tony dying, though, and let it go down.

Ultimately, the Scarlet Witch seemingly sacrificed herself when she destroyed Wundagore Castle, leaving everyone to believe that she died in the rubble. Despite that, a hidden detail in the sequel's ending may hint that Wanda is still alive.

Clea asks if her beloved is all right, to which Strange responds "No. I am dead." Strange survived the mortal blow - but only by dying and being resurrected by Death in the same instant, officially killing him for a fourth time.
Black Bolt could have killed the Scarlet Witch in Doctor Strange: Multiverse of Madness if he had attacked immediately, before Reed Richards talked. Likewise, Maria Rambeau, the Captain Marvel of that universe, could have killed the Scarlet Witch if she had attacked the moment Black Bolt died.
