Immortal tribrids can be killed via decapitation and heart extraction. If an immortal tribrid is bound and deprived of their werewolf nature they can then burn to death in the sunlight with a daylight amulet.
Wood: A wooden stake through the heart will desiccate a tribrid and temporarily "kill" them. However, wood from what appears to be a Red Oak tree can permanently destroy a tribrid as Nature created a balance near the site of the tribrids' birth through the sapling that Hope, Lizzie, and Josie created.
Created by Hope, Lizzie, and Josie, it is unknowingly one of Nature's loopholes that finds balance with the birth of the tribrid, Hope Mikaelson, and its wood is seemingly the only thing that can kill her.
Recently reborn, Lizzie is only a Newborn vampire and has not got control of her powers or humanity yet whereas, as a Tribrid, Hope has the powers of a werewolf. This means that, even if Hope hadn't accidentally bonded Lizzie to her in the transformation, the Heretic wouldn't have been able to kill the Tribrid.

“The Story of My Life” reveals that Lizzie had not just taken any random vampire's blood but had consumed Hope's blood, leaving Lizzie with a sire bond to Hope. This effectively ends Lizzie's ability to kill her, as the sire bond prevents Lizzie from inflicting fatal harm upon the Tribrid.
About Hope Mikaelson:
She is the daughter of Niklaus Mikaelson and Hayley Marshall-Kenner, which makes her the only tribrid on the series. Hope Mikaelson is the only character that is hybrid of all three bloodlines: werewolf, witch, and vampire.
Due to the lack of blood flow, tribrids do not have any bodily functions. Because of this, they do not produce supplication and are unable to become pregnant but they can impregnate female humans.

Klaus is around 1000 years old, he was transformed into a vampire during the Viking Age, by his mother Esther. It was also during that time he turned into a werewolf for the first time. So, he has been a hybrid for around 1000 year. A physical fight, Hope can never win.
For pretty much all of Klaus' vampire life, Mikael was the only man who was physically capable of fighting Klaus, who actually had the drive and determination to kill him, and most importantly had the only weapon that could actually kill Klaus.
