Alison's parents (as far as we know, anyway) are Jessica and Kenneth DiLaurentis. Jason (Drew Van Acker) is her biological half-brother, as they share Jessica as a mother.
Kenneth DiLaurentis is the father of Alison DiLaurentis and the legal father of Jason DiLaurentis who comes off as intimidating and angry at first. He first appeared in Crazy and was played by Jim Abele. He has never forgiven Hanna for saying she saw Alison three days before she was discovered dead.
Her birth mother, Mary Drake, was a patient at Radley while she was pregnant. Jessica, Mary's sister, told Veronica Hastings that Mary was pregnant with Veronica's husband, Peter Hastings's child. Veronica couldn't stand the thought of the child being forgotten, so she agreed to adopt her.
Wren is the father.
In the series finale, it was revealed that Wren is the father — but Emily and Alison don't appear to know that information.
Peter Hastings is the father of Melissa, Jason, Spencer, and Alex Drake. He had an affair with Jessica DiLaurentis which resulted in the birth of Jason DiLaurentis. He also had a one night stand/affair with Mary Drake (who he thought was her twin sister, Jessica), which resulted in the birth of both Spencer and Alex.
Charlotte was shocked to see Spencer's twin, though the two became close overtime and called each other sisters. Wren and Alex fell in love, and Wren proceeded to work with Charlotte during her time as A. He later helped Alex when she decided to take over the game and avenge Charlotte's death.
Melissa was unaware of Spencer's adoption and thought they were biological sisters. They are actually half-sisters, as they share the same father but different mothers. They are also adopted sisters since Veronica adopted Spencer. They share a half-brother, Jason DiLaurentis.

He is mad at Alison for lying to him about her age.
Mary then cradles Spencer's body and reveals that she is indeed Spencer's mother, making Spencer the biological sister of Cece Drake.
The big reveal, of course, was the identity of Uber A. It turns out that Spencer (Troian Bellisario) had an evil twin this entire time, and that "A.D." refers to her initials — Alex Drake.
The scene where Veronica revealed all of this to Spencer was heartbreaking. In her mind, there was no doubt that Spencer was her child even though they were not biologically related; she loved Spencer and would protect her at all costs. But all Spencer could see were the lies she had been told her entire life.
A turned out to be CeCe Drake, the adopted sister of Alison DiLaurentis, who went missing and was presumed dead by the end of the show's pilot episode.
Officially introduced in the series finale, Alex Drake is both the identical twin of main character Spencer Hastings and the previously anonymous identity known as "A.D." In childhood, Drake was adopted by a wealthy British family that later abandoned her, forcing Alex to bounce around foster homes and orphanages.
Courtney DiLaurentis was the identical twin sister of Alison DiLaurentis. She was also the girl who Aria, Spencer, Emily, and Hanna knew as "Ali" and was their best friend. She was actually not mentally insane, Alison just made a plan to make Courtney look like it.
Lily DiLaurentis-Fields is the daughter of Emily Fields and Alison DiLaurentis. She is also the twin sister of Grace DiLaurentis-Fields. She was born during the time jump in "'Til Death Do Us Part".
Emily and Alison fell in love after Alison was artificially inseminated by Archer; she was actually carrying babies made with Emily's eggs, and the two got together and eventually got married, raising their twin daughters.
The official endgame couples are: Hanna Marin is married to Caleb Rivers. Aria Montgomery is married to Ezra Fitz. Emily Fields was engaged to Alison DiLaurentis in the finale.
It was revealed in "Free Fall" that he had a brief relationship with Alison DiLaurentis and has been tracking her and The Liars in order to write a "true crime novel" and figure out who 'A' is.

According to her taped confession — which you know “A” is going to get his/her/its hands on — Melissa saw Spencer with a shovel the night Alison disappeared. She thought Spencer killed Ali, so she pushed her into a ditch and buried her.
Although Melissa does not believe Spencer at first that her husband is dead, she starts to wonder if Spencer might know something that she doesn't, reasoning that Ian would never leave his pregnant wife.
Charlotte knew that Spencer was her half-sister prior to the Dollhouse, but viewed Spencer as a doll and not a sister. They share Jason DiLaurentis as a brother - Spencer is Jason's paternal half-sister while Charlotte is Jason's adoptive sister.
