It is revealed that she was killed because she interrupted the demon as he fed his blood to baby Sam. Sam learns for the first time that he was fed demon blood, and that Mary knew the demon that killed her.
33 years later, after Dean reunited God and The Darkness, she was resurrected by the latter as a gift for Dean.
Castiel attempts to travel to Heaven to find Mary's soul for a resurrection while Jack approaches Rowena to use a spell from the Book of the Damned to resurrect Mary himself. Despite Rowena's warnings as Mary left no body behind, Jack attempts the spell, but only succeeds in bringing back Mary's body.
Jack says he snapped and it was over quickly. He wishes it didn't happen. Dean is barely holding it together. He doesn't like that Jack is calling Mary's death, “the accident.” But he plays along and says they understand and they forgive Jack.

Mary Winchester was the first death in the Supernatural TV series; however, in season eleven of the show, she was brought back to life by Amara, The Darkness. She was reconnected with her sons, Sam and Dean, and began aiding them in the family business, saving people and hunting things.
Jack subsequently regains his powers after killing Michael and absorbing his grace. However, killing Michael burns away a great deal of Jack's soul, leaving him soulless.

Cassandra Masters is the daughter of Castiel and Meg Masters and the only known Nephalem to exist, which is a hybrid between a union of an angel and demon, even being part human due of Castiel and Meg's human vessels. Cassandra acts as one of the leading main characters with Sam and Dean.
Dean appears visibly saddened by Castiel's lack of faith in him. Castiel later returns in Two Minutes to Midnight and apologizes to Dean for not having more faith in him. Dean forgives Cass, and they reconcile.

John secretly summons Azazel and seems to know what the demon's plans are. He then makes a deal to save Dean, giving up his life, soul, and the Colt. Before dying, John tells Dean, if he can't save Sam, he'll have to kill him, should he become evil.
After Jack helped to save his colony, Bobby fully reconciled with Mary. During Let the Good Times Roll, the two walk together and share a talk with bobby remarking he may want to stay in the new universe even if it would take time to adjust.
